NOMID (Neonatal-Onset Multisystem Inflammatory Disease) is a sporadic disorder affecting approximately 100 people worldwide. What are the FDA-Approved Treatments for NOMID?

 

NOMID, also known as CINCA (Chronic Infantile Neurological Cutaneous and Articular syndrome), is mainly caused by CIAS1/NLRP3 gene mutation, resulting in an increased activity of interleukin (IL) 1ß. Anakinra and Canakinumab are the only drugs approved by the FDA to treat NOMID.

 

  • Anakinra : It is a recombinant human interleukin-1 receptor antagonist (IL-1Ra) that blocks the production of interleukin by binding to the receptor. This medication is available as a subcutaneous injection that needs to be administered once daily. It has a half-life of 4-6 hours and helps manage symptoms such as brain inflammation, fever, joint pain, vision loss, hearing loss, and rash.  
  • Canakinumab : It falls under the same drug class as anakinra (interleukin-1 inhibitor), but has a different mechanism of action. It is also approved by the FDA for treating NOMID. The drug is a monoclonal antibody that targets IL-1β (interleukin) and stops its activity altogether. It has a half-life of 26 days and is given every 2 months.
